Degeneracy of the Antithrombin Binding Sequence in Heparin: 2-O-Sulfated Iduronic Acid Can Replace the Critical Glucuronic Acid

Heparin binds to and activates antithrombin (AT) through a specific pentasaccharide sequence, in which a trisaccharide subsite, containing glucuronic acid (GlcA), has been considered as the initiator in the recognition of the polysaccharide by the protein. Recently it was suggested that sulfated iduronic acid (IdoA2S) could replace this “canonical” GlcA. Indeed, a heparin octasaccharidic sequence obtained by chemoenzymatic synthesis, in which GlcA is replaced with IdoA2S, has been found to similarly bind to and activate antithrombin. By using saturation-transfer-difference (STD) NMR, NOEs, transferred NOEs (tr-NOEs) NMR and molecular dynamics, we show that, upon binding to AT, this IdoA2S unit develops comparable interactions with AT as GlcA. Interestingly, two IdoA2S units, both present in a ¹C₄-²S₀ equilibrium in the unbound saccharide, shift to full ²S₀ and full ¹C₄ upon binding to antithrombin, providing the best illustration of the critical role of iduronic acid conformational flexibility in biological systems.     

Effects of intramolecular hydrogen bonds on phosphorescence emission: A theoretical perspective

Importing intramolecular hydrogen bond in phosphorescent transition metal complexes has been considered one of the excellent approaches to improve the electroluminescence performance of organic light-emitting diodes in real applications. However, the relationships between such H-bond structure and phosphorescent properties have not been theoretically revealed yet. In this study, two types of intramolecular hydrogen bonds are introduced into the two classes of traditional materials, that is, Pt(II) and Ir(III) complexes ( 1a and 2a ) to completely elucidate their influence on the structures and properties by comparing with the original phosphors ( 1b and 2b ) using density functional theory/time-dependent density functional theory for the first time. A comprehensive analysis of the geometric structures, molecular orbitals, and luminescence properties (including phosphorescence emission wavelengths and radiative and nonradiative decay processes) has been carried out. Our theoretical model highlights that complexes 1a and 2a embedded with H-bonds significantly promote the phosphorescence emission band blue-shifted and restrict molecular deformations compared with the corresponding 1b and 2b , which can provide helpful guidance to regulate and design several aspects of highly efficient blue phosphorescent emitters.

单相氟磷灰石微晶玻璃晶相特征和生物相容性研究

采用熔融-淬火-烧结法制备氟磷灰石微晶玻璃,研究其析晶机制和体外生物相容性.采用X射线衍射(XRD)和场发射扫描电镜(FE-SEM)分析了不同烧结温度下样品的晶相组成和微观结构,利用能量色散X射线光谱仪(EDS)分析了化学成分.结果表明,氟磷灰石是烧结微晶玻璃中的唯一晶相,其晶体可以在较宽的温度范围内析出.显微组织结构致密,显微硬度为644～709 Hv0.1.氟磷灰石晶体在FE-SEM中有针状和多边形两种形态,多边形是短棒状晶体的横断面.低温下,以固相烧结为主,氟磷灰石析晶以表面析晶为主,晶体生长为针形;高温下玻璃液较多,在保温和冷却凝固过程中析出短棒状晶.在37℃温度下将样品在模拟体液中浸泡14 d,SEM/EDS和FT-IR检测结果表明,样品表面包含磷酸根和羟基的吸收带,形成了羟基磷灰石层,可作为骨或牙齿修复材料.

Permanent antimicrobial silicone rubber based on bonding guanidine polymers

Recently, silicone rubber (VMQ) was extensively used in household articles and medical devices. To develop a kind of safe and long‐term antimicrobial VMQ was of great significance. In this work, a kind of vinyl‐contained polyhexamethylene guanidine hydrochloride (VPHMG) was synthesized and used as antimicrobial additive for VMQ. With the increasing of VPHMG addition, the mechanical properties and antimicrobial properties of VMQ‐VPHMG were significantly improved. In particular, the antimicrobial rates against Escherichia coli and Staphylococcus aureus were higher than 99.99% as for 4 wt% of VPHMG addition. Moreover, the surface concentration of VPHMG as well as the antimicrobial rates revealed almost unchanged after being extracted by water and methanol. All the results indicated the vinyl‐contained VPHMG vulcanization and therefore provided the permanent antimicrobial performance for VMQ.

Highly stable polyoxometalate‐resorcin[4]arene‐based inorganic‐organic complexes for catalytic oxidation desulfurization

Self‐assembly of a resorcin[4]arene‐based ligand (TMR4A) with metal salts and H₃PMo₁₂O₄₀·xH₂O offers two isostructural complexes, namely, [Ni₂Cl(TMR4A)₂(CH₃CN)₂]·[PMo₁₂O₄₀]·4CH₃CN ( 1 ) and [Co₂Cl(TMR4A)₂(CH₃CN)₂]·[PMo₁₂O₄₀]·4CH₃CN ( 2 ). In both 1 and 2 , one Cl^? anion bridges two metal cations, and each metal cation is further chelated by four 2‐mercaptopyridine N‐oxide groups of one TMR4A, producing a [M₂Cl(TMR4A)₂]³⁺ dimer (M = Ni or Co). The negative [PMo₁₂O₄₀]^3? as a counter‐anion balances the positive charge. Markedly, 1 and 2 exhibit high stability in aqueous solutions with different pH values and in organic solvents. Remarkably, the efficient heterogeneous catalytic capability for oxidative desulfurization was studied by suing 1 and 2 as recycled catalysts. Moreover, the electrochemical behaviors of the two compounds were discussed as well.
